Optimize your cloning: No more trouble with ligation

Abstract: One of the most critical steps in molecular cloning is ligation, the joining of DNA fragments driven by T4 DNA Ligase. Intact and compatible DNA ends are an important pre-requisite to make this reaction successful. But T4 DNA Ligase itself has several special features and preferences which, if taken into consideration, can help to improve ligation efficiency tremendously. With this webinar we will give an overview on different DNA end modifications, how to set up an optimal ligation reaction and how to assess and improve ligation efficiency. Also, we will discuss alternatives to traditional T4 DNA Ligase based cloning, such as PCR cloning and LIC.

Regina studied biology, followed by a Ph.D. in Microbioly and Post doc in Physiology. She joined Thermo Scientific 2.5 years ago as a technical support specialist. During this time she deepened her knowledge on cloning techniques, PCR and qPCR as well as other molecular biology applications.
